Summary

The Center for Breakthrough Medicines is seeking a talented and passionate Senior/Associate Principal Scientist to join its Analytical Development, Next-Generation Sequencing (NGS) team. As a Senior/Associate Principal Scientist, you will provide expertise in NGS-based analyses of DNA and RNA, while contributing to the development, optimization, and validation of clinical-grade assays to measure complex, blood-based analytes (e.g., whole-genome sequencing of cfDNA). You will work collaboratively with multiple teams to develop and optimize the assays in a regulated, high-throughput environment as well as identify ways to rapidly iterate and improve on existing methods to evaluate technologies that can enhance our end-to-end test performance. The successful candidate must be able to set and understand priorities in a local and matrixed environment, must be a team player with a collaborative spirit, and capable of building and maintaining networks within and outside the organization. 

Responsibilities

  • Responsible for NGS wet lab and dry lab assay development, validation and transfer utilizing Illumina, PacBio and Oxford Nanopore platforms.
  • Improve cost, efficiency, and performance of existing wet and dry lab workflows
  • Research scientific literature and apply current and emerging NGS methods for advanced characterization of gene and cell therapy biologicals and outcomes of gene editing experiments.
  • Communicate and collaborate with R&D, Process Development and Manufacturing teams to identify NGS applications within current and future processes.
  • Develop, optimize, and validate NGS library preparation, quality control and sequencing methodologies.
  • Provide wet lab support to deliver custom and routine NGS projects.
  • Adapt new and existing NGS methods for high-throughput automation and liquid handling.
  • Perform primary NGS data analysis and data manipulation on Windows and Unix platforms.

Qualifications

  • PhD in biological sciences with 1-3 years of experience or Master’s/Bachelor’s degree with 5-10 years of experience required.
  • Deep understanding of molecular biology principles and proficiency in molecular biology procedures is required.
  • Direct experience with molecular biology assay development is required.
  • Knowledge of NGS applications for characterization of viral vectors, molecular cloning, gene transfer and gene editing is highly desired.
  • Practical knowledge of Windows molecular biology software (SnapGene, Vector NTI, IGV, USCS Genome Browser etc). Knowledge of Unix command line is a plus.
  • Understanding of advanced NGS techniques such as different types of RNA sequencing, chromatin sequencing, targeted resequencing, custom primer optimization, single cell technologies is a plus.
  • Understanding of modern vectorology, AAV and lentivirus production is a plus.
  • Ability to develop, improve and meticulously follow test methods and standard operating procedures (SOPs) in GxP environment.
  • Highly collaborative mindset, strong interpersonal skills, excellent verbal and written communication skills.
